DOSIS EKSTRAK BIJI KOPI Ekstrak biji kopi hijau diberikan dengan dosis 400 mg/kgBB. Dosis ini merupakan dosis yang berpotensi anti-obesitas (Shimoda, Seki, & Aitani, 2006).

Penetapan Kadar Kafein dengan Spektrofoto-metri UV/Vis (Cunniff 1995) Pembuatan kurva kalibrasi dilakukan dengan membuat larutan kafein pada rentang konsentrasi 4-9 ppm menggunakan pelarut kloroform dan diukur pada absorbansi maksimum pada λ 275 nm. Serbuk kopi dibasakan dengan NH4OH, dipanaskan pada suhu 100°C selama 2 menit, didinginkan, dilarutkan dalam kloroform, dan disaring. Filtrat diukur absorbansinya menggunakan spektofotometri UV/Vis pada λ 275 nm. Raw coffee beans were ground and screened through a 250 µm sieve to get a uniform texture. An accurately weighed amount of sieved coffee and green tea leaf powder (approximately 50 mg each) was dissolved in 25 ml of distilled water. The solution was stirred for one hour using a magnetic stirrer and heated gently to remove caffeine easily from the solution. Then, the solution was filtered using a glass filter to get rid of the particles.
