• Modeling & simulation in drug discovery and development • PK principles: steady state, exposure, iv vs. extravascular dosing • PD principles: kinetics of drug action, effect compartment models, turnover delays Statistical issues in modeling: weighting, model discrimination, goodness of it, • combining results from various subjects, destructive sampling strategies and population sampling designs • PK/PD modeling using WinNonlin’s newly released version 6 • Strategies for dose selection for first-in-man studies Hands-on practical exercises: one-compartment bolus iv, PK/PD modeling using an effect compartment, • simulation of time course to select optimal dose and regimen, modeling pharmacodynamic time delays, case studies on the value of modeling and simulation
