EXERCICE D’APPLICATION DE CALCUL D’ACTIVITE ENZYMATIQUE : Ex 1 : Etude de la cinétique de la βgalactosidase.

Ex 2 : Calcul d’activité catalytique

1. Retracer le graphique correspondant au résultat expérimental, A =f (t) et schématiser le contenu du tube expérimental en rappelant les volumes et les dilutions éventuelles.

2. Rappeler la loi de Beer Lambert et exprimer Vi

3. A partir de Vi calculer l’activité enzymatique Z, attention à la dilution de l’enzyme !

4. En déduire b la concentration d’activité catalytique « Cat c »

5. Calculer Zsp en µmolde S hydrolyé/min/mg de protéine

CORRECTION EXO 1 : -

Z = Vmax x VR

Vi = Vmax = 12 µmol / mL / min Z = 12 x 1 = 12 UI = 200 nkat -

Catc = Z / Eez = Vmax x (VR /VE) = 200/0,2 = 1 µkat/mL

AS = Z / mPn = 1 /0,2 = 5 nkat/mg de Pn

EXO 2

Vi= V mx = Δ C / Δ t = (0.3 / 5000) x (5/3) = 0.0001 mol/L/min = 100 u /L de MR

Z = ( Vm x (VR / VE)) x f = ((0.0001 x ( 3 / 0,1 )) x 500 = 1,5 mol/L

donc 1500 µmol de S hydrolyse /ml soit 1500 U/mL d’extrait -

Zsp = 1500 / 3 = 500 U /mg
